Directions For Use
Allow test device, urine specimen, and/or controls to equilibrate to room
temperature (15-30°C) prior to testing.
Bring the pouch to room temperature before opening it. Remove the test device
from the sealed pouch and use it as soon as possible.
Place the test device on a clean and level surface. Hold the dropper vertically and
transfer 3 full drops of urine (approx. 100μl) to the specimen well (S) of the test device, and
then start the timer. Avoid trapping air bubbles in the specimen well (S). Refer to illustration.
Wait for the red line(s) to appear. The result should be read at 5 minutes. Results may be
stable up to 4 hours after test initiation.
INTERPRETATION OF RESULTS
NEGATIVE:* Two lines appear. One red line should be in the control region (C), and
another apparent red or pink line should be in the test region (T). This negative result
indicates that the Oxycodone concentration is below the detectable level (100 ng/mL).
* NOTE: The shade of red in the test region (T) may vary, but it should be considered
negative whenever there is even a faint pink line.
POSITIVE: One red line appears in the control region (C). No line appears in the
test region. This positive result indicates that the Oxycodone concentration is above
the detectable level (100 ng/mL).
INVALID: Control line fails to appear. Insufficient specimen volume or
incorrect procedural techniques are the most likely reasons for control line
failure. Review the procedure and repeat the test using a new test device. If the
problem persists, discontinue using the lot immediately and contact your local
distributor.

SUMMARY
In drug analysis, oral fluid drug screening has certain advantages over the screening of other biological fluids such as urine and blood. Oral fluid is readily accessible, and is less likely to be adulterated. With oral fluid tests, the drugs may be detected immediately after ingestion, even before they are metabolized and would show up in urine. In general, OraLine® IV is designed to work at a lower detection level for all test drugs than those detected in urine samples. OraLine® IV oral fluid screening for drugs of abuse detects the presence of parent compounds and drug metabolites. The OraLine® IV uses an immunochromatographic technique that provides the accuracy of an immunoassay without the need for laboratory equipment . The concentrations of the drugs or drug metabolites are at levels greater than or equal to the cutoff value of the assay specified on the attached table. The immunoreaction causes a displacement in the antibody-antigen complex that is visually detected on a chromatographic membrane. The test is designed to use a fresh oral fluid sample directly, and to provide rapid test results for on-site testing purposes . The OraLine® IV detects recent drug usage, and is particularly useful for screening of drug intoxication related to any impairment situations.

Cocaine is a potent CNS stimulant and a local anesthetic derived from the coca plant (erythroxylum coca).
The drug is often self-administered by nasal inhalation, intravenous injection

of 40 ng/mL, codeine can be detected in the oral fluid within 1 hour following a single oral dose and can
remain detectable for 7-21 hours after the dose.2 6-Monoacetylmorphine (6-MAM) is found more prevalently
in oral fluid, and is a metabolic product of heroin. Morphine is a major metabolic product of codeine and
heroin, and is detectable for 24-48 hours following an opiate dose.

the morphine concentration in oral fluid exceeds 40 ng/mL.
Marijuana (THC)
Historical studies have shown a window of detection for THC in oral fluid of up to 14 hours
after drug use.3
The THC assay contained within the OrALert TM Oral Fluid Drug Screen Device yields a positive result when the
Delta9-THC concentration in oral fluid exceeds 100 ng/mL.
Methamphetamine (mAMP)
Depending on the route of administration, methamphetamine can be detected in oral fluid as early
as 5-10 minutes following use.1 Methamphetamine can be detected in oral fluid for up to 72 hours after use.

result when the methamphetamine concentration in oral fluid exceeds 50 ng/mL.

INTERPRETATION OF RESULTS
The test results are determined by the presence or absence of red lines. With a POSITIVE sample, the drug or drug metabolite from the
sample solution blocks the limited antibody binding sites on the colored microspheres. This reaction prevents the attachment of the colored
microspheres to the antigenic probe sites on the membrane. Therefore, a POSITIVE test result for one of the four drugs is indicated by the
absence of its corresponding red line. A NEGATIVE test result for all of the four drugs is indicated by the presence of all five (5) red lines.
Lines MET, OPI, COC, and THC are the drug probe lines, and line C is the control or reference line (see diagrams). Line C is not related to
the presence of any drug, but shows that the microsphere particles have completely traveled along the membrane and that the test is viable.
The line intensity may vary for each drug. Do not compare line intensities when interpreting results.
The OraLine® IV is a qualitative test and the intensity of the lines does not correspond to drug concentration. The OraLine® IV only
indicates the presumptive presence or absence of a specific drug in the oral fluid sample, and provides preliminary qualitative test results.
Below are examples of a few possible test results (more variations are possible):
NOTE: Interpret the test results after the control line C is clearly defined in the test window. No test results should be interpreted until
the control line C can be clearly identified in the viewing area. In the event that the control line C does not form, the test results are
inconclusive and the test should be repeated using a new OraLine® IV device.

Allow test device, urine specimen, and/or controls to equilibrate to room
temperature (15-30°C) prior to testing.
Bring the pouch to room temperature before opening it. Remove the test device
from the sealed pouch and use it as soon as possible.
Place the test device on a clean and level surface. Hold the dropper vertically and
transfer 3 full drops of urine (approx. 100μl) to the specimen well (S) of the test device, and
then start the timer. Avoid trapping air bubbles in the specimen well (S). Refer to illustration.
Wait for the red line(s) to appear. The result should be read at 5 minutes. Results may be
stable up to 4 hours after test initiation.
INTERPRETATION OF RESULTS
NEGATIVE:* Two lines appear. One red line should be in the control region (C), and
another apparent red or pink line should be in the test region (T). This negative result
indicates that the Oxycodone concentration is below the detectable level (100 ng/mL).
* NOTE: The shade of red in the test region (T) may vary, but it should be considered
negative whenever there is even a faint pink line.
POSITIVE: One red line appears in the control region (C). No line appears in the
test region. This positive result indicates that the Oxycodone concentration is above
the detectable level (100 ng/mL).
INVALID: Control line fails to appear. Insufficient specimen volume or
incorrect procedural techniques are the most likely reasons for control line
failure. Review the procedure and repeat the test using a new test device. If the
problem persists, discontinue using the lot immediately and contact your local
distributor.

SUMMARY
The OrALert Oral Fluid Drug Screen Device for AMP mAMP COC OPI THC PCP and their metabolites is a rapid, oral fluid screening test that can be performed without the use of an instrument. The test utilizes monoclonal antibodies to selectively detect elevated levels of specific drugs in human oral fluid.
Amphetamine (AMP)
Depending on the route of administration, amphetamine can be detected in oral fluid as early as 5-10 minutes following use.1 Amphetamine can be detected in oral fluid for up to 72
hours after use.
The Amphetamine assay contained within the OrALert TM Oral Fluid Drug Screen Device yields a positive result
when the amphetamine concentration in oral fluid exceeds 50 ng/mL.
Cocaine (COC)
Cocaine is a potent CNS stimulant and a local anesthetic derived from the coca plant (erythroxylum coca).
The drug is often self-administered by nasal inhalation, intravenous injection
Opiates (OPI)
Using an immunoassay cutoff level
of 40 ng/mL, codeine can be detected in the oral fluid within 1 hour following a single oral dose and can
remain detectable for 7-21 hours after the dose.2 6-Monoacetylmorphine (6-MAM) is found more prevalently
in oral fluid, and is a metabolic product of heroin. Morphine is a major metabolic product of codeine and
heroin, and is detectable for 24-48 hours following an opiate dose.
The Opiates assay contained within the OrALert TM Oral Fluid Drug Screen Device yields a positive result when
the morphine concentration in oral fluid exceeds 40 ng/mL.
Marijuana (THC)
Historical studies have shown a window of detection for THC in oral fluid of up to 14 hours
after drug use.3
The THC assay contained within the OrALert TM Oral Fluid Drug Screen Device yields a positive result when the
Delta9-THC concentration in oral fluid exceeds 100 ng/mL.
Methamphetamine (mAMP)

Phencyclidine (PCP)
Phencyclidine (PCP), the hallucinogen commonly referred to as Angel Dust, can be detected in oral fluid as a
result of the exchange of the drug between the circulatory system and the oral cavity. In a paired serum and
oral fluid sample collection of 100 patients in a hospital emergency department, PCP was detected in the oral
fluid of 79 patients at levels as low as 2 ng/mL and as high as 600 ng/mL.
The Phencyclidine assay contained within the OrALert TM Oral Fluid Drug Screen Device yields a positive result
when the PCP concentration in oral fluid exceeds 10 ng/mL.

The OrALert TM Oral Fluid Drug Screen Device for AMP/mAMP/COC/OPI/THC/PCP is an immunoassay based on the principle of competitive binding. Drugs that may be present in the oral fluid specimen compete against their respective drug conjugates for binding sites on their specific antibody.
During testing, a portion of the oral fluid specimen migrates upward by capillary action. A drug, if present in the oral fluid specimen below its cut-off concentration, will not saturate the binding sites of its specific
antibody. The antibody will then react with the drug-protein conjugate and a visible colored line will show up in the test line region of the specific drug strip. The presence of drug above the cut-off concentration in the oral
fluid specimen will saturate all the binding sites of the antibody. Therefore, the colored line will not form in the test line region.
A drug-positive oral fluid specimen will not generate a colored line in the specific test line region of the strip because of drug competition, while a drug-negative oral fluid specimen will generate a line in the test line
region because of the absence of drug competition.
To serve as a procedural control, a colored line will always appear at the control line region, indicating that proper volume of specimen has been added and membrane wicking has occurred.

S.V.T. SUMMARY
The Adulteeration strip contains chemically treated reagent pads. 3-5 minutes following the activation of the reagent pads by the urine sample, the colors that appear on the pads can be compared with the printed color
chart card. The color comparison provides a semi-quantitative screen for any combination of oxidants/pyridinium chlorochromate (PCC), specific gravity, pH, nitrite, glutaraldehyde and creatinine in human urine which can help assess the integrity of the urine sample.
What is Adulteeration?
Adulteration is the tampering of a urine specimen with the intention of altering the test results. The use of adulterants can cause false negative results in drug tests by either interfering with the screening test and/or destroying the drugs present in the urine. Dilution may also be employed in an attempt to produce false negative drug test results.
One of the best ways to test for adulteration or dilution is to determine certain urinary characteristics such as pH and specific gravity and to detect the presence of oxidants/PCC, specific gravity, pH, nitrite, glutaraldehyde and creatinine in urine.
Oxidants/PCC (Pyridinium chlorochromate) tests for the presence of oxidizing agents such as bleach and hydrogen peroxide. Pyridinium chlorochromate (sold under the brand name UrineLuck) is a commonly used adulterant.8 Normal human urine should not contain oxidants or PCC.
Specific gravity? tests for sample dilution. The normal range is from 1.003 to 1.030. Values outside this range may be the result of specimen dilution or adulteration.
pH tests for the presence of acidic or alkaline adulterants in urine. Normal pH levels should be in the range of 4.0 to 9.0. Values outside of this range may indicate the sample has been altered.
Nitrite tests for commonly used commercial adulterants such as Klear or Whizzies. They work by oxidizing the major cannabinoid metabolite THC-COOH.9 Normal urine should contain no trace of nitrite. Positive results generally indicate the presence of an adulterant.
Glutaraldehyde tests for the presence of an aldehyde. Adulterants such as UrinAid and Clear Choice contain glutaraldehyde which may cause false negative screening results by disrupting the enzyme used in some immunoassay tests.8 Glutaraldehyde is not normally found in urine; therefore, detection of glutaraldehyde in a urine specimen is generally an indicator of adulteration.
Creatinine is a waste product of creatine; an amino-acid contained in muscle tissue and found in urine.2 A person may attempt to foil a test by drinking excessive amounts of water or diuretics such as herbal teas to flush the system. Creatinine and specific gravity are two ways to check for dilution and flushing, which are the most common mechanisms used in an attempt to circumvent drug testing. Low creatinine and specific gravity levels may indicate dilute urine. The absence of creatinine (< 5 mg/dl) is indicative of a specimen not consistent with human urine.
The Multi-Drug 10 Panel Test is an all inclusive, point of use screening test for the rapid detection of 11-nor-Δ9-Tetrahydrocannabinol-9-carboxylic acid (THC/Marijuana), Cocaine and its metabolite, benzoylecgonine, PCP (Phencyclidine), Morphine and its related metabolites derived from opium (opiates), Methamphetamines (including Ecstasy*), Methadone, Amphetamines, Barbiturates, Benzodiazepines and Tricyclic Antidepressants (TCA) in human urine at or above the system concentrations levels established as standard minimums by the National Institute on Drug Abuse (NIDA), the World Health Organization (WHO) and SAMHSA as shown in the chart to the right. Note system cut-off concentrations are expressed in nanograms per milliliter solution.

Allow test device, urine specimen, and/or controls to equilibrate to room
temperature (15-30°C) prior to testing.
Bring the pouch to room temperature before opening it. Remove the test device
from the sealed pouch and use it as soon as possible.
Place the test device on a clean and level surface. Hold the dropper vertically and
transfer 3 full drops of urine (approx. 100μl) to the specimen well (S) of the test device, and
then start the timer. Avoid trapping air bubbles in the specimen well (S). Refer to illustration.
Wait for the red line(s) to appear. The result should be read at 5 minutes. Results may be
stable up to 4 hours after test initiation.
INTERPRETATION OF RESULTS
NEGATIVE:* Two lines appear. One red line should be in the control region (C), and
another apparent red or pink line should be in the test region (T). This negative result
indicates that the Oxycodone concentration is below the detectable level (100 ng/mL).
* NOTE: The shade of red in the test region (T) may vary, but it should be considered
negative whenever there is even a faint pink line.
POSITIVE: One red line appears in the control region (C). No line appears in the
test region. This positive result indicates that the Oxycodone concentration is above
the detectable level (100 ng/mL).
INVALID: Control line fails to appear. Insufficient specimen volume or
incorrect procedural techniques are the most likely reasons for control line
failure. Review the procedure and repeat the test using a new test device. If the
problem persists, discontinue using the lot immediately and contact your local
distributor.
